Formation of organic iodides from containment paint ingredients caused by gamma irradiation

The formation of volatile alkyl iodides other than methyl iodide during a serious nuclear reactor accident may have radiological significance. The hypothesis that radioactive alkyl iodides, other than methyl iodide, could form from paint solvents under the conditions of a serious nuclear accident in light water reactors (under boiling water reactor (BWR) and pressurised water reactor (PWR) conditions) was tested using stable elemental iodine, a gamma irradiator and gas chromatography equipment. It was found that methyl and isopropyl iodides were formed from the texanol ester, which is used in many modern water-based paints. Methyl, ethyl, propyl and butyl iodides were formed from a hydrocarbon solvent (white spirit) commonly used in paint products used in the past. These results suggest that further work on the formation and behaviour of the higher alkyl iodides (containing more than one carbon atom) under the conditions of a serious nuclear accident is justified.

Iodide excess regulates its own efflux: a possible involvement of pendrin

Adequate iodide supply and metabolism are essential for thyroid hormones synthesis. In thyrocytes, iodide uptake is mediated by the sodium-iodide symporter, but several proteins appear to be involved in iodide efflux. Previous studies demonstrated that pendrin is able to mediate apical efflux of iodide in thyrocytes. Acute iodide excess transiently impairs thyroid hormone synthesis, a phenomenon known as the Wolff-Chaikoff effect. Although the escape from this inhibitory effect is not completely understood, it has been related to the inhibition of sodium-iodide symporter-mediated iodide uptake. However, the effects of iodide excess on iodide efflux have not been characterized. Herein, we investigated the consequences of iodide excess on pendrin abundance, subcellular localization, and iodide efflux in rat thyroid PCC13 cells. Our results indicate that iodide excess increases pendrin abundance and plasma membrane insertion after 24 h of treatment. Moreover, iodide excess increases pendrin half-life. Finally, iodide exposure also increases iodide efflux from PCC13 cells. In conclusion, these data suggest that pendrin may have an important role in mediating iodide efflux in thyrocytes, especially under conditions of iodide excess.

Enhanced iodide sequestration by 3-biphenyl-5,6-dihydroimidazo[2,1-b]thiazole in sodium/iodide symporter (NIS)-expressing cells

The ability of the sodium/iodide symporter (NIS) to take up iodide has long provided the basis for cytoreductive gene therapy and cancer treatment with radioiodide. One of the major limitations of this approach is that radioiodide retention in NIS-expressing cells is not sufficient for their destruction. We identified and characterized a small organic molecule capable of increasing iodide retention in HEK293 cells permanently transfected with human NIS cDNA (hNIS-HEK293) and in the rat thyroid-derived cell line FRTL-5. In the presence of 3-biphenyl-4′-yl-5,6-dihydroimidazo[2,1-b)thiazole (ISA1), the transmembrane iodide concentration gradient was increased up to 4.5-fold. Our experiments indicate that the imidazothiazole derivative acts either by inhibiting anion efflux mechanisms, or by promoting the relocation of iodide into subcellular compartments. This new compound is not only an attractive chemical tool to investigate the mechanisms of iodide flux at the cellular level, but also opens promising perspectives in the treatment of cancer after NIS gene transfer.

Measurements by microcalorimetry and diffuse reflectance spectroscopy have pointed out the nucleation and growth stages during J-aggregation of a cyanine dye on cubic AgBr emulsion crystals treated with iodide. For the cyanine dye studied, the integral molar adsorption enthalpy on an iodide-doped AgBr surface is higher than that on the pure AgBr surface. J-aggregation is observed on AgBr crystals having a surface iodide concentration greater-than-or-equal-to 16 atom%. Below this iodide concentration, only dye monomers and dimers are formed at dye coverage less-than-or-equal-to 0.4. Differential adsorption enthalpy measurements versus dye coverage reveal the influence of dye-dye interaction on the adsorption enthalpy. The presence of iodide on AgBr influences the nucleation stage of J-aggregate formation. XPS and XAES experiments show that the Ag 3d5/2-I 3d5/2 binding energy of surface iodide at low iodide concentrations is significantly larger than that of surface AgI or of surface AgBrI. The chemical state of this surface iodide is interpreted to be influenced by high-energy sites located near corners and edges of the cubic AgBr grains, as revealed by imaging SIMS experiments.
