Two consecutive Eley-Rideal reaction steps in a D atom adsorbate interaction: from methylene iodide via methyl iodide to methane

The interaction of gaseous D atoms with methylene iodide molecules adsorbed on graphite monolayer covered Pt(lll) surfaces was studied using direct product rate measurement. At 85 K methane was observed as a reaction product from two consecutive D/adsorbate reaction steps. Via D addition to methylene iodide, adsorbed methyl iodide is formed as an intermediate. Subsequently, D abstracts methyl from methyl iodide towards methane. Approaching and passing the methyl iodide intermediate desorption temperature of 135 K during the reaction, the desorption of the intermediate prevents methane production. The kinetics of methane and intermediate methyl iodide formation is in agreement with the operation of Eley-Rideal reaction steps. This is in accordance with the expectation that on a substrate with a flat adsorption well for atoms from the gas phase the Eley Rideal mechanism should operate in atom/adsorbate reactions. The cellular expression of the sodium iodide symporter (NIS) has been shown to confer iodide-concentrating capacity in non-thyroid cell types. We examined the role of NIS in the uptake of the alpha-particle emitting radiohalide [At-211]astatide in the UVW human glioma cell line transfected to express NIS. [At-211]Astatide uptake is shown to be NIS-dependent, with characteristics similar to [I-131]iodide uptake. These studies suggest [At-211]astatide as a possible alternative radionuclide to [I-131]iodide for NIS-based endoradiotherapy, and provide a model for the study of [At-211]astatide behavior at a cellular level. INHIBITION EFFECT OF POTASSIUM-IODIDE ON CORROSION OF STAINLESS-STEEL IN HYDROCHLORIC-ACID SOLUTION

The inhibition effect of potassium iodide on the corrosion of stainless steel has been examined using weight loss and electrochemical measurements. Potassium iodide was found to be an excellent inhibitor for stainless steel types 316, 430 and 440 (efficiency more than 95%), and moderate for type 304. The corrosion rates in 1 M HCl and in presence of potassium iodide fit an Arrhenius equation. The presence of potassium iodide lowered both the activation energy and the rate of reaction. Polarization measurements indicated that potassium iodide acts as an anodic inhibitor particularly at higher concentrations and as a cathodic inhibitor at lower concentrations. The synergistic effect of potassium iodide on the inhibitive efficiency of the organic substances was studied.

Expression of the human sodium/iodide symporter (hNIS) in xenotransplanted human thyroid carcinoma

The uptake of iodide in thyroid epithelial cells is mediated by the sodium/iodide symporter (NIS). The uptake of iodide is of vital importance for thyroid physiology and is a prerequisite for radioiodine therapy in thyroid cancer. Loss of iodide uptake due to diminished expression of the human NIS (hNIS) is frequently observed in metastasized thyroid cancer. So far, no animal model for the study of radioiodine therapy in thyroid cancer has been available. Strategies to restore iodide uptake in thyroid cancer include the exploration of hNIS gene transfer into hNIS defective thyroid cancer. We have performed a stable transfection of hNIS into the hNIS defective follicular thyroid carcinoma cell line FTC133. Stabily transfected colonies exhibited high uptake of (NaI)-I-125, which could be blocked completely with sodiumperchlorate. hNIS transfected FTC133 and non-transfected cell lines injected subcutaneously in nude mice formed tumors after 6 weeks. Iodide uptake in the hNIS transfected tumor was much higher than in non-transfected tumor, but a rapid release of radioactivity from the hNIS transfected tumor was observed. Further studies are necessary to investigate the role of hNIS in relation to other thyroid specific proteins in iodide metabolism in thyroid cancer.

Study of potential inhibitors of thyroid iodide uptake by using CHO cells stably expressing the human sodium/iodide symporter (hNIS) protein

Background: Thyroid gland is highly dependent on dietary intake of iodine for normal function, so it is particularly subjected to endocrine disruptor action. The human sodium/iodide symporter (hNIS) is an integral plasma membrane glycoprotein mediating the active transport of iodide into thyroid follicular cells, a crucial step for thyroid hormone biosynthesis. Beyond to perchlorate and thyocianate ions a few other inhibitors of iodide uptake have been described. Aim: The aim of this study was to investigate if 10 substances usually used as drugs in clinical practice were able to inhibit NIS-mediated iodide uptake in vitro. Materials and methods: A CHO cell line stably expressing hNIS was used test any inhibition of NIS-mediated iodide uptake exerted by drugs. Perchlorate and thyocianate ions were used as positive controls. Results: None of the analyzed substances was able to significantly inhibit iodide uptake in our system. As we expected, perchlorate and thyocianate ions were able to inhibit iodide uptake in a dose-dependent manner. Conclusions: In conclusion, we carried out an in vitro assay to evaluate the potential inhibitory effect of common drugs on NIS-mediated iodide uptake by using CHO-hNIS cells. None of the analyzed substances was able to inhibit iodide uptake; only perchlorate and thyocianate were able to inhibit iodide uptake in a dose-dependent manner. (J. Endocrinol.
